I do not have much experience with AWS Instances, but I assume that has no influence to the general procedure.
I'm not sure if you did the normal upgrade-path from version 2012 to 2016/16 ?
You have to install 2015/16 ( regardless if SQL Express or SQL) completele fresh with a fresh new dB ( on your SQL ) and thereafter you can start migrating the old data from 2012 with the upgrade wizard !
For that choose "Environment" then click on "Upgrade of 2012 Environment" and follow the instructions.
