In current versions you should see the reason for that - in 2018 not always - so it's not easy to check...
Try another user from this client to check if it could be a firewall / network problem - if other user could connect - check the permissions on SQL again for that user...

So, if keep same backend (SQL server 2016 + AD group for permission), can I directly upgrade this user app to current version (ASG RD 2020)?

The user will not be able to work with the old data source without upgrading - but the connection test should run before - so just try to install version 2020 side by side - and try to connect to the old database - perhaps we can see the detailed error then
